Inside the DNA Lab (What Really Happens)

After scientists at the lab receive your DNA sample, collected from your easy and painless cheek swab, they gently extract your genetic material.

To extract your DNA, CRI scientists add enzymes and detergents to a solution that cause those cells to pop open and release DNA. This process is called “lysis.”

Then they add alcohol to the solution, which helps separate the released DNA from the rest of the cellular debris in your sample. This is a process called “precipitation,” as the DNA clumps together and rises into the alcohol.

Once your DNA is successfully extracted and isolated, CRI’s scientists put it through a rigorous inspection. They want to ensure enough DNA survived the journey, so they can create your personalized package of detailed genetic insights.

How Advanced Genotyping Reveals Your Genetic Blueprint

CRI’s laboratory utilizes the cutting-edge Illumina iScan system, enabling a profound and comprehensive exploration of your DNA.

When your DNA is successfully extracted and passes inspection, it gets bonded to a “genotyping chip.” This chip then goes through the Illumina iScan system, which blasts the chip with lasers.

These lasers cause the DNA to glow in different colors to represent genetic variations, which then get scanned into raw data.

The building blocks of DNA are chemical bases called nucleotides, which come in four varieties — A, T, C, and G (adenine, thymine, cytosine, and guanine, respectively).

You have 3 billion pairs of these bases, so 6 billion letters in all, strung together in a sequence. Altogether, this genetic information is called your genome.

Based on the glowing colors present on the genotyping chip, the computers determine which of the four letters is present at the 527,414 relevant locations in your genome.

For example, one “SNP” near the HERC2 gene codes for a protein believed to be involved with melanin production. Melanin is a pigment responsible for skin, eye, and hair colors. Which letter — or genetic variation — you have on this relevant SNP could play a direct role in your skin, eye, and hair color.
